Adenosine deaminase (ADA)

Adenosine deaminase ADA (EC:3.5.4.4) is an important enzyme in the metabolism of purine nucleoside. It belongs to the mercaptoprotein enzyme. Each molecule contains at least 2 active mercaptocyls, and its activity can completely inhibit chloramalgic acid. ADA can catalyze the transformation of adenine nucleoside into subxanthine nucleoside, and then generate subxanthine through the action of nucleoside phosphorylase. The final product of metabolism alleviation is uric acid. ADA is widely distributed in various tissues of the human body, with the highest content in thymus, spleen and other lymphoid tissues, and lower content in liver, lung, kidney and bone muscles. In the blood, ADA mainly exists in red blood cells, granulocytes and lymphocytes.
Its activity is about 40 to 70 times that of serum. Human ADA gene is located on the 20th chromosome and presents genetic polymorphism. There are three isozyme, ADA1, ADA1+CP and ADA2 respectively.ADA1 has a molecular weight of 35kD and is a single chain protein. The molecular weight of ADA1+CP is 280kD, which is actually formed by the combination of two single bonds of ADA1 molecules and a human non-enzymatic binding protein of 200kD. The molecular weight of ADA2 is 100kD. The three isozyme have different dynamic properties: the optimal PH of ADA1 and ADA1+CP is 5.5 ~ 8.0, and the Km value of adenosine is 50umol/L. The ratio of 2 '-deoxyadenosine to adenosine activity was 0.8, both of which were sensitive to the inhibition of erythro-9- (2-hydroxy-3-nonyl) adenine (EHNA). Compared with the first two isozyme, ADA2 has a lower optimal PH, a higher catalytic adenosine Km value (2000umol/L) and a lower ratio of 2 '-deoxyadenosine to adenosine activity (0.2), which is not sensitive to the inhibition of EHNA. The distribution of ADA isozyme in various tissues and cells is different. ADA1 is dominant in spleen, RBC, lymphocytes, monocytes and neutrophils. ADA1+CP is dominant in liver, lung, pancreas, kidney tissue and bone.
